Separation and biophysical characterisation of macromolecules
The separation and biophysical characterisation of macromolecules plays a key role in understanding the molecular weight, size distribution and structural properties of biomolecules in solution. This technical sheet describes the use of size‑based separation techniques combined with multi‑detector analysis to achieve absolute characterisation. Multi‑Angle Light Scattering (MALS) is applied after Size‑Exclusion Chromatography (SEC) or Asymmetric Flow Field‑Flow Fractionation (AF4) to determine molecular weight distribution and size‑related parameters. AF4, a widely used Field Flow Fractionation technique, enables the separation of macromolecules and nanoparticles across a broad size range while requiring limited sample preparation.
