Development of cell-based potency assays for mRNA-LNPs using flow cytometry and Gyrolab
Lipid nanoparticle formulations have become the preferred delivery system for mRNA therapeutics. As their biological activity depends on efficient cellular delivery and protein expression, reliable potency assays are essential to assess product performance throughout development.
This poster highlights the development of two complementary cell-based approaches for mRNA-LNP potency testing. Flow cytometry is used to quantify the proportion of cells expressing the target protein following transfection, while Gyrolab immunoassays directly measure protein expression levels. The study also evaluates dose-response relationships, repeatability, intermediate precision, and the comparability of both platforms for potency determination.
By combining cellular and protein-based readouts, these orthogonal methods provide a robust and flexible strategy for assessing mRNA-LNP potency and supporting the development of mRNA therapeutics.
